This is a working overview of CJC-1295, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-12-11. Anything still debated is marked as such rather than presented as settled.
Two forms circulate in research settings and are frequently confused. One carries the drug affinity complex and is often written as CJC-1295 with DAC; the other lacks that group and is usually called modified GRF(1-29). The two share the same core sequence but differ sharply in how long they persist in blood. Products labelled only as CJC-1295 normally refer to the version carrying the complex. Documentation that omits the distinction leaves the intended molecule ambiguous.
CJC-1295 is a synthetic peptide built as a long-acting analogue of growth hormone-releasing hormone. Its backbone matches the first twenty-nine residues of the natural human hormone, with four amino acid substitutions added to slow enzymatic breakdown. A reactive maleimide group, commonly termed the drug affinity complex, allows the peptide to attach to circulating albumin after administration. That albumin attachment keeps the molecule in the bloodstream for an extended period instead of being cleared within minutes.
The compound emerged from work at a Canadian biotechnology firm in the early 2000s. Early human studies examined its effect on growth hormone and insulin-like growth factor 1 in healthy volunteers and in people with HIV-associated fat redistribution. Reports described sustained increases in both markers after a single injection. Development did not advance to regulatory approval, and the clinical programme was later discontinued. The molecule is now encountered mainly as a research chemical rather than a marketed medicine.
Four amino acid substitutions separate the modified backbone from the parent GRF(1-29) sequence. These changes reduce recognition by dipeptidyl peptidase IV and related proteases, extending the interval before degradation. Development work in this area sought longer-acting GHRH analogs for endocrine investigation. Published descriptions treat the substitution set as a defining property of the core sequence, while the albumin-binding linker is described separately as an optional addition to that same backbone.
CJC-1295 is a synthetic peptide analog of growth hormone-releasing hormone, constructed on the 29-amino-acid fragment designated GRF(1-29). The name began as an internal development code during the 1990s and later spread through research supply catalogs and discussion forums. The molecule does not occur in nature; its sequence is engineered rather than isolated from tissue. Two related compounds are sold under this single label, and they differ by one appended chemical group that strongly influences how long the peptide remains in circulation.

Batch-to-batch consistency depends on solid-phase peptide synthesis and subsequent purification. Coupling efficiency, resin choice, and cleavage conditions all affect the final profile. Counter-ion content and moisture can shift the apparent mass of a batch. Documentation typically includes a certificate of analysis with chromatograms and spectra. Independent verification by a second laboratory is sometimes requested. Whether a given certificate reflects the actual vial contents depends on chain of custody. Analytical methods themselves carry uncertainty that should be stated alongside results.
Lyophilized material is typically stored at minus twenty degrees Celsius or lower. Keeping the vial dry and protected from light preserves peptide integrity. Repeated freeze-thaw cycles can cause aggregation or loss of activity. Once dissolved, solutions are generally kept at two to eight degrees Celsius. Stability data for reconstituted solutions vary, and long-term behavior is not fully established. Working aliquots reduce the number of times a stock container is opened.
Reverse-phase high-performance liquid chromatography is the standard tool for purity assessment. The technique separates the target peptide from truncated or modified byproducts. Mass spectrometry confirms molecular weight and supports sequence verification. Electrospray ionization and matrix-assisted laser desorption are both used. Amino acid analysis provides an independent check on composition. Purity values are commonly reported as area percentage from the chromatogram. Residual trifluoroacetate and water content are also measured in many quality programs.
Laboratory handling centers on minimizing exposure to water, heat and oxygen before use. Working solutions are typically prepared in sterile water or a mild buffer, and any residual particulate matter is removed by filtration. When the powder dissolves slowly, a small proportion of acetonitrile or dilute acetic acid is sometimes added as a co-solvent. Containers are kept sealed and desiccated between uses. Records of lot number, reconstitution date and storage conditions support later comparison of results across experiments.
Identity and purity are established with reversed phase high performance liquid chromatography coupled to mass spectrometry. The chromatographic step separates the target peptide from truncated sequences and deletion products, while the mass measurement confirms the expected molecular weight to within a fraction of a dalton. Because the two common variants differ by the presence of the linker, mass alone can distinguish them in the unconjugated state. Amino acid analysis and peptide mapping are used when sequence level confirmation is required.
Lyophilized material is generally stable for extended periods when held at minus twenty degrees Celsius or below and protected from moisture and light. In solution the peptide is more labile; bond hydrolysis, aggregation and oxidation of susceptible residues all proceed faster at ambient temperature. Repeated freeze and thaw cycles should be avoided because they promote clumping and loss of soluble material. The conjugated variant adds a further consideration, since the maleimide group can hydrolyze in aqueous buffer and lose its ability to react with albumin.
==== Recombinant DNA ==== Biotechnology in the modern sense of genetic engineering began in the 1970s, with the invention of recombinant DNA techniques. Restriction enzymes were discovered and characterized in the late 1960s, following on the heels of the isolation, then duplication, then synthesis of viral genes. Beginning with the lab of Paul Berg in 1972 (aided by EcoRI from Herbert Boyer's lab, building on work with ligase by Arthur Kornberg's lab), molecular biologists put these pieces together to produce the first transgenic organisms. Soon after, others began using plasmid vectors and adding genes for antibiotic resistance, greatly increasing the reach of the recombinant techniques. Wary of the potential dangers (particularly the possibility of a prolific bacteria with a viral cancer-causing gene), the scientific community as well as a wide range of scientific outsiders reacted to these developments with both enthusiasm and fearful restraint. Prominent molecular biologists led by Berg suggested a temporary moratorium on recombinant DNA research until the dangers could be assessed and policies could be created. This moratorium was largely respected, until the participants in the 1975 Asilomar Conference on Recombinant DNA created policy recommendations and concluded that the technology could be used safely. Following Asilomar, new genetic engineering techniques and applications developed rapidly. DNA sequencing methods improved greatly (pioneered by Frederick Sanger and Walter Gilbert), as did oligonucleotide synthesis and transfection techniques.
=== Lipid metabolism === PLP is an essential component of enzymes that facilitate the biosynthesis of sphingolipids. Particularly, the synthesis of ceramide requires PLP. In this reaction, serine is decarboxylated and combined with palmitoyl-CoA to form sphinganine, which is combined with a fatty acyl-CoA to form dihydroceramide. This compound is then further desaturated to form ceramide. In addition, the breakdown of sphingolipids is also dependent on vitamin B6 because sphingosine-1-phosphate lyase, the enzyme responsible for breaking down sphingosine-1-phosphate, is also PLP-dependent.

=== Oliver and Schäfer (1894) === British physician George Oliver and physiologist Edward Albert Schäfer, professor at University College London, collaborated on the physiological effects of adrenal extracts. They first published their findings in two reports in 1894, a full publication followed in 1895. Though frequently falsely attributed to secretin, found in 1902 by Bayliss and Starling, Oliver and Schäfer's adrenal extract containing adrenaline, the substance causing the physiological changes, was the first hormone to be discovered. The term hormone would later be coined by Starling.

Moroidin is one of several biologically active compounds isolated from the venom of Dendrocnide moroides, a member of the stinging nettle family. The plant stores its venom in silica hairs that break off when touched, delivering the toxins through the skin and inducing extreme pain. Moroidin also produces a similar pain response when injected subdermally, so it is thought to be partially responsible for the plant’s toxicity. However, moroidin injections are not as potent as injections of crude matter isolated from Dendrocnide moroides, suggesting that there are additional stinging toxins in the venom.
== Genetics == In terms of the genetics of UCMD1, there are mutations in the genes COL6A1, COL6A2, and COL6A3. This sub-type of muscular dystrophy is both autosomal recessive and autosomal dominant in nature. COL6A1 plays an important part in maintaining the human body's integrity of various tissues. Alpha 1 subunit of type VI collagen is the encoded protein. In terms of the genetics of UCMD2, there are mutations in the gene COL12A1, and is autosomal recessive. Collagen VI is a principal structural component of the extracellular matrix (ECM), where it self-assembles into a distinct microfibrillar network that links the muscle fiber's basal lamina to the surrounding interstitial matrix, thereby anchoring muscle cells to the ECM. Mutations in COL6A1, COL6A2, and COL6A3 impair the proper assembly, secretion, or incorporation of collagen VI microfibrils into the ECM, disrupting its structural integrity and contributing to the muscle-fiber degeneration observed in UCMD.

=== Other appearances === Culp appeared in the 1993 live action video game Voyeur as the game's villain, industrialist/politician Reed Hawke. He lent his voice to the digital character Dr. Wallace Breen, the prime antagonist in the 2004 computer game Half-Life 2. The video clip of "Guilty Conscience" features Culp as an erudite and detached narrator describing the scenes where Eminem and Dr. Dre rap lyrics against each other. He only appears in the music video. In the album version, the narrator is Mark Avery. On November 9, 2007, on The O'Reilly Factor, host Bill O'Reilly interviewed Culp about the actor's career and awarded Culp with the distinction "TV Icon of the Week".
Currently approved CCEEVs are also more thermotolerant than the label suggests: a commercial CCEEV packaged as a ready-to-inject solution in a vial, with a 2–8 °C recommendation from the manufacturer, was found to have lost none of its effectiveness in dogs after spending three months at 30 °C (86 °F). It is believed that liquid vaccines degrade by processes such as aggregation of protein particles, so dry human vaccines are expected to be even more stable. Indeed, a human CCEEV was found to remain effective after three months at 37 °C (99 °F).

=== Chinese Civil War: relations with Nationalists and Communists === Liu Wenhui's relationship with the Nationalist government continued to be one of grudging acceptance due to the fact that Xikang was too far from the central government in Nanjing to reliably influence or coerce. It is also believed by historian Kim Hee-shin that Chiang tolerated Liu's presence in Xikang to prevent the Sichuan warlord Liu Xiang from becoming too powerful. In 1936 Liu Wenhui's ties with Chiang soured even further due to his independent policy, but Chiang was not powerful enough to do anything meaningful against him at the time. Liu's relationship had a cooperative relationship with Sichuan governor Zhang Qun, the leader of the KMT's Political Science Clique. Both men being Sichuan natives, Liu was able to appeal to these provincial ties to enlist Zhang's support in negotiations with Chiang. Zhang, for his part, supported Liu as a counterweight to other Sichuanese warlords. In 1939, fighting broke out in Garzê between the officers of the deceased Panchen Lama and Liu Wenhui's army. Liu spread rumors about northern Kham potentially being granted to the Panchen Lama's office by the Kuomintang, and drove out the Lama's men. Although the Panchen Lama's forces were normally KMT-aligned, the central government remained on good terms with Liu Wenhui, who by now had taken advantage of the war against Japan to acquire more funding for Xikang. In January 1947, an anti-Liu uprising led by Zhu Shizheng (朱世正) captured the towns of Lushan and Tianqian and attempted to take Ya'an.
Stanley L. Inhorn (August 1, 1928 – February 19, 2025) was an American physician who was professor of Pathology and Laboratory Medicine at the University of Wisconsin-Madison, where he also served as Director of the Wisconsin State Laboratory of Hygiene (WSLH) on the UW campus. A graduate of Cleveland's Western Reserve University and Columbia University Medical School, Inhorn moved to Madison in 1953 to pursue an internship and residency in pathology before being appointed Assistant Professor of Pathology and Assistant Director of the WSLH in 1960. He became Director of the WSLH in 1966, a position he held until 1979, when he was asked by the UW Medical School to create a Department of Pathology and Laboratory Medicine.
=== RNaseA === In 1959, Fred Richards discovered that a proteolytic enzyme, Subtilisin, had the ability to cleave the enzyme Ribonuclease A into two components, a peptide corresponding to the first 20 amino acids of the enzyme (S-Peptide) and the remainder of the protein (S-Protein). When separated from one another, each piece was inactive but when they were simply mixed together, full enzymatic activity was restored. Hofmann speculated that this system might be a model for the way peptide hormones interact with their receptors. Structure-function studies with ACTH were complicated by the necessity to assess activity in the whole animal. The S-Peptide:S-Protein system afforded a simple system with none of the biological complications inherent in testing ACTH analogs. To study which amino acids might be important in establishing the binding between peptide hormones and their receptors, Hofmann and his group began a systematic evaluation of the contributions each amino acid in the S-Peptide molecule made to the binding with S-Protein. The ability of synthetic analogs of S-Peptide to activate S-Protein correlated well with those of synthetic ACTH analogs to elicit hormonal activity: 1) only a portion of the S-Peptide chain was essential for re-establishing full activity with the S-Protein; 2) methionine was not important; 3) substituting one particular amino acid in the peptide not only destroyed the activity of the peptide but created an antagonist as well.
=== Postmortem and forensic === After death, the vitreous resists putrefaction longer than other body fluids. Within the hours, days and weeks after death, the vitreous potassium concentration rises, at such a predictable speed that vitreous potassium levels are frequently used to estimate the time since death (post-mortem interval) of a corpse. The metabolic exchange and equilibration between systemic circulation and vitreous humour is so slow that vitreous humour is sometimes the fluid of choice for postmortem analysis of glucose levels or substances which would be more rapidly diffused, degraded, excreted or metabolized from general circulation. According to Judaism, extracting the vitreous fluid for forensic chemical analysis is preferred to blood analysis (in case a forensic or post-mortem toxicology test is deemed necessary). This avoids the loss of even a few droplets of blood from the body prior to burial.
== Diagnosis == The disease may be diagnosed based on a clinical examination, which identifies signs and symptoms generally associated with the people who have the condition. Genetic testing for known pathological variants is preferred, by testing of the COL6A1, COL6A2, COL6A3 and COL12A1 genes. In the case of a VUS, testing of dermal fibroblast culture is used for an accurate diagnosis. Additional laboratory tests may be performed before genetic testing, such as creatine kinase (CK) blood test, MRI of the muscles, and electromyography (EMG). Phenotypes of overlap between Ullrich congenital muscular dystrophy (UCMD) and Bethlem can be assumed. In the differential diagnosis of UCMD, even in patients without finger contractures, Bethlem myopathy could be considered.

It is a synthetic peptide analogue of growth hormone-releasing hormone. Four substitutions in its sequence make it more resistant to enzymatic degradation than the natural hormone. In the version carrying a drug affinity complex, the peptide binds albumin and remains in circulation for days.
No regulatory agency has approved CJC-1295 for clinical use. Human trials were conducted in the 2000s, but the development programme was discontinued before any marketing application succeeded. Material sold today is offered as a research chemical, and its purity depends on the supplier.
Sermorelin is an unmodified fragment of growth hormone-releasing hormone and is cleared quickly. CJC-1295 contains substitutions that resist breakdown, and the form with a drug affinity complex persists much longer. Both act at the same receptor but differ substantially in duration of action.
No. It is a laboratory-synthesized analog built on a fragment of the natural hormone. The natural peptide is shorter-lived and lacks the stabilizing substitutions found in the synthetic version.
